Description
Thermostable Inorganic Pyrophosphatase catalyzes the hydrolysis of inorganic pyrophosphate to form orthophosphate: P2O7-4 + H2O -> 2HPO4-2. This enzyme has 100% activity when heated at 100°C for 4 h, so it can be used in PCR reaction to remove the inhibition of inorganic pyrophosphate on PCR and enhance DNA replication.
Unit Definition
One unit is the amount of enzyme that will generate 1 μmol of phosphate per minute from inorganic pyrophosphate under standard reaction conditions (a 10 minute reaction at 75°C in 50 mM Tricine [pH 8.5] , 1 mM MgCl2, 0.32 mM PPi, reaction volume of 0.5 ml).
Note
lThe enzyme is active in a variety of reaction buffers. Therefore, it can be directly used in tHDA amplification, PCR and sequencing.
lThe dosage of the enzyme needs to be optimized in different experiments, usually in the concentration of 5 - 50U/ml.
lThe enzyme could not be inactivated by heat.
